Polymerase chain reaction, or PCR, is a technique to make many copies of a specific DNA region in vitro and relies on a thermostable DNA polymerase, Taq polymerase, and requires DNA primers designed specifically for the DNA region of interest. Using PCR it is possible to generate thousands to millions of copies of a particular section of DNA from a very small amount of DNA and is a common tool that is routinely used in DNA cloning, medical diagnostics, and forensic analysis of DNA. PCR technique is based on the natural processes a cell uses to replicate a new DNA strand. Five core ‘ingredients’ are required to set up a PCR i.e. the DNA template to be copied, primers, DNA nucleotide bases, Taq polymerase enzyme, and buffer. PCR involves a process of heating and cooling called thermal cycling which is carried out by a machine in three main stages Denaturing, Annealing and Extending. These three stages are repeated 20-40 times, doubling the number of DNA copies each time. After PCR has been completed, electrophoresis can be used to check the quantity and size of the DNA fragments produced. PCR is used in many areas of biology and medicine, including molecular biology research, medical diagnostics, and even some branches of ecology.
